This review explains that gelatin capsule shells cross-link when exposed to aldehydes or to high humidity and temperature, forming near-permanent covalent bonds between lysine side chains that slow release, and that the United States Pharmacopeia therefore adopted a two-tier dissolution test adding pepsin below pH 6.8 or pancreatin above it when a capsule fails on the first tier.
